Activated liquid thromboplastin is a rabbit brain extract, which is highly sensitive to coagulation factors II, V, VII and of calcium chloride 4.2M separately.

The reagent bottle can remain stable for 12 months if kept refrigerated between 2 and 8 C. Once the bottle is opened, the reagent can be used until finished without losing its activity, as long as it is kept refrigerated, not has been preheated, does not become contaminated and does not freeze.

Test time and temperature

Check the test time in the insert

1. All material used in the test must be washed with double-distilled water and used exclusively for the determination of prothrombin time.

2. The mixture of anticoagulant and blood must be exact.

3. The reagents must reach the optimal temperature (37ºC) and heating the test plasma for more than 5 min must be avoided.

4. Be careful not to contaminate the thromboplastin bottle with plasma or calcium chloride.

5. If heating blocks are used, they conduct heat more slowly, so the time for them to reach the optimal temperature may be longer.

6. Whenever the prothrombin time test is done, a normal and abnormal control should be run.

7. All elements of the system must be maintained to work optimally.

8. A quality control program is recommended in order to keep under control all the variables that may influence the results.
